PhD - Microbiome and Psoriasis

The Laboratory of Cellular and Molecular Immunology MBU AS CR is looking for a highly motivated PhD student to collaborate in the project focusing on the role of the microbiome in the pathogenesis of psoriasis. This project has significant implications for clinical medicine. The main aim of your work would be to distinguish subtypes of patients with psoriasis based on clinical characteristics, molecular signatures and the specific composition of their microbiota. This characterisation will help to predict the success of the biological therapy and thus has potential to tailor the appropriate therapy.

Post-doctoral Position open at Inserm U1184

The Autoimmunity Research Team is seeking passionate and self-motivated post-doctoral candidates to embark on research endeavors focusing on the role of RNA methylation in mice models of Sjogren disease (SjD). The position is currently available and funded for 2 years, subject to renewal.

Sjögren disease (SjD) is a prototypic systemic autoimmune disease affecting 0.1% of thepopulation and characterized by the dysfunction of salivary and lacrimal glands, along with numerous systemic complications and an increased risk of lymphoma. For a long timeorphaned by treatment, there is now a revolution under way, with several phase II and phase III trials in the field.

Environment: Our team, led by Pr Xavier Mariette - within Inserm UMR 1184, CEA, Université Paris-Saclay, is dedicated to exploring the pathophysiology, epidemiology, and treatment strategies for various autoimmune conditions. Our research encompasses rheumatoid
arthritis, SjD and lymphomas associated with autoimmune diseases, among other autoimmune-related disorders. We are committed to advancing our understanding of these complex diseases, investigating the underlying mechanisms driving their progression, and identifying novel therapeutic interventions. Through multidisciplinary approaches and collaborative efforts, we aim to make significant strides in improving patient outcomes and quality of life for individuals affected by autoimmune disorders. The laboratory provides state-of-the-art facilities for cellular and molecular biology, and top-level rheumatology/immunology research and an animal facility. The host lab has a particular interest in translating its basic science discoveries into translational research.

Candidates: The applicants are expected to have a strong background in immunology and immunopathology (PhD), RNA biology and cellular biology as well as in mice work, including injections and organ recuperation. The ideal candidates should be highly motivated, curious, and enthusiastic to work in a highly collaborating team. Proven ability to identify research objectives and meet agreed deadlines, self-motivation, flexibility, and assistance to other ongoing research work is essential. Excellent written and communication skills in English are required. Bioinformatics and international training will constitute an advantage.

Assistant Professor in Immunology, Paris-Saclay

Start date 1st September 2024. 
TEACHING :  
The assistant professor will integrate the Immunology teaching team of the Faculty of Pharmacy Paris-Saclay. He/she will take part in the teaching of fundamental immunology and clinical immunology given in the 2nd and 3rd years of the common basic training in pharmacy. He/she will also be involved in supervising tutored projects, as well as in several teaching units (of the "Biology applied to therapeutic and diagnostic innovation" and "Immunology" master programs at the University of Paris-Saclay, for which the department is responsible. The assistant professor may be required to teach in English in the immunology courses of the international "Development of Drugs and Health Products" master's program. Some of the lectures may be remote or hybrid teaching.  Teaching is provided using a variety of methods, including flipped teaching, project-based training and the use of active teaching tools (e.g. wooclap). After his/her recruitment, the assistant professor will

RESEARCH :  
The assistant professor will join the "Immunoregulation, chemokines and viral persistence" team in the  INSERM  UMR996  unit  (http://umr996.inserm.fr/teams/team-i/),  located  in  the  Henri  Moissan 
building  (faculty  of  pharmacy  Paris-Saclay).  The  team  focuses  on  epithelial  immunity  and  human papillomavirus (HPV) host interactions. These viruses are known to cause 5% of human cancers, but all individuals are healthy carriers of HPV in their microbiota and only a minority of infections lead to pathogenesis.  
In this context, the team is characterizing the mechanisms by which HPV replication and pathogenesis are controlled within pluristratified epithelia, and the impact of these viruses on the local immune fitness,  based  on  data  from  both  healthy  subjects  and  patients  suffering  from  primary immunodeficiencies associated with exacerbated HPV pathogenesis (e.g. WHIM syndrome due to a gain of function of the CXCL12/CXCR4 chemokine/receptor axis). The focus is on epithelial immune 
cells,  particularly  dendritic  cells,  and  their  interactions  with  keratinocytes  in  the  context  of asymptomatic infection or HPV pathogenesis. The effects of certain candidate molecules for targeted therapies are also studied.  

This work is being carried out using patient samples, three-dimensional (3D) epithelial cell cultures and original transgenic mouse models, as part of national and European collaborations funded by several programs (e.g. Marie Curie actions, Fondation pour la Recherche Médicale, Agence Nationale pour la Recherche).
The assistant professor will draw on his/her expertise to develop a project that will include one or more of the following areas: 
- characterization of the interactions between keratinocytes and immune cells during HPV replication or pathogenesis   
- understanding the mechanisms by which the CXCL12 axis and its receptors CXCR4 and ACKR3, or other factors, regulate epithelial immunity 
He/she will be able to draw on the host team's expertise in immunology, virology and cell biology and in cutting-edge methodological approaches (e.g. 3D cultures, mass/spectral cytometry, mass imaging, RNAseq, etc.), as well as on the supportive environment of the Henri Moissan site. 

The project developed by the lecturer recruited to the team will be in line with the Graduate School's flagship  themes  of  "Health  and  Drug  Sciences"  and  the  interdisciplinary  themes  of  "HEALth  and THerapeutic Innovation" and "Microbes" in particular, enabling access to local calls for tenders to obtain funding for transdisciplinary projects
