Phd Position - Neutrophil-driven Pulmonary disorders-doctoral positionPost-doctoral position

NOVEL REGULATION PATHWAYS OF NEUTROPHIL SERINE PROTEASE (NSP) ACTIVATION

In chronic lung diseases such as Cystic Fibrosis lung disease or Chronic Obstructive Pulmonary Disease, the lung is susceptible to destruction of lung tissue by human cells that normally participate in removing bacteria from the lung. These human cells (neutrophils) release enzymes that can degrade lung tissue and cause inflammation in the lung. In this study, we will evaluate how these enzymes are synthesised prior to their release from neutrophils which will include development of new inhibitors to regulate their activity

Neutrophil Serine Proteases (NSPs) such as Neutrophil Elastase (NE) are known to be present in the lungs of individuals with various chronic lung diseases including Cystic Fibrosis (CF) lung disease and Chronic Obstructive Pulmonary Disease (COPD). It is known that NE correlates significantly to the development of bronchiectasis in early CF and other studies have demonstrated that inhibition of another protease, cathepsin C (CatC), which is known to regulate NSP activation, results in reduced frequency of exacerbation in individuals with bronchiectasis. However, recent data from the Taggart laboratory shows that there is another level of regulation of NSP activity. The proposed project will delineate further this novel NSP processing activity and evaluate how this activity impacts neutrophil function including oxidant generation and bacterial killing. In addition, there will be a focus on measurement of protease activity assays and analysis of neutrophil gene expression. The study will be a collaboration between the Taggart laboratory in the Wellcome Wolfson Institute for Experimental Medicine, the Williams laboratory in the Patrick G Johnston Centre for Cancer Research, the Tirouvanziam laboratory in the Emory University, Atlanta, USA and the company partner ProAxsis, where the student will spend time conducting protease activity assays.

Postdoc position – Immunosuppressive factors in the tumor microenvironment as modulators of immunotherapy efficacy

We are seeking a highly motivated candidate for a postdoc position within the field of cancer immunology. The applicant must have a background in cancer biology or immunology. Prior in vivo research expertise is an advantage.

Keywords: Tumor microenvironment, immunotherapy, mouse models, hypoxia, extracellular matrix, T cell suppression

The research group: The project will be carried out in the Tumor Stroma and Matrix Immunology (TSMI) group at the National Center for Cancer Immunotherapy (CCIT-DK), Department of Oncology, Copenhagen University Hospital, Herlev, Denmark, a research center with a strong background in experimental and clinical cancer immunotherapy research. Many clinical immunotherapy trials are currently running at CCIT, several of which originate from our own pre-clinical research results. The TSMI group is one of five research groups at the center and is headed by Associate Professor and Lundbeck Fellow, Daniel H. Madsen. The goal of the research group is to discover molecular mechanisms by which the tumor microenvironment influences immunotherapy efficacy, and to use this knowledge for the development of new therapeutic strategies for improving immunotherapy. The TSMI group is currently studying how the extracellular matrix and different cellular components of the tumor microenvironment affect the activity of tumor-infiltrating T cells. We also focus on the role of hypoxia for immunotherapy efficacy and on the discovery of new targets for improving cancer immunotherapy. The research projects are among other things based on mouse models, 3D cell culture assays, and clinical samples from cancer patients. The TSMI group is an international group currently consisting of two postdocs, three PhD students, and two Master’s students.

The research project: Cancer immunotherapy has had a major breakthrough within the last decade with the discoveries that endogenous cytotoxic T cells possess the ability to react against cancer cells and that the blocking of immunosuppressive signals can be sufficient for obtaining extremely impressive clinical results. Still, however, many cancer patients do not respond to immunotherapy. One likely reason for this is that the local T cell response in the tumor gets suppressed by immunosuppressive factors in the tumor microenvironment.
The selected candidate will work on a project within the focus area of the TSMI group. The exact research project will be defined after discussions with the applicant, taking into consideration the interests and expertise of the applicant. The project will involve methods such as mouse cancer models, 3D cell culture assays, RNA-sequencing, flow cytometry, immunohistochemistry, co-culture assays, cytokine profiling, and FACS-isolation of specific cell populations for functional assays.

Responsibilities:

  • Conduct in vivo and in vitro experiments
  • Independently plan and advance the research projects
  • Collaborate with internal and external partners
  • Communicate research results
  • Contribute to the overall work of the group to achieve common goals. This includes discussions and strategic planning of projects within the group.
  • Involvement in administrative and practical tasks required for the laboratory work to proceed optimally.
Qualifications:

  • A PhD within the field of cancer biology/immunology
  • Experience with relevant techniques and, preferably, with in vivo research with mice
  • Fluency in English
As a person, you are a motivated researcher driven by scientific curiosity. You are capable of solving problems independently, but you also possess good team spirit and excellent communication skills.

Start date: May 1st, 2024 or after agreement.
Duration: 3 years

Application procedure:
Application deadline is February 15th, 2024.
The application should include a motivated letter of application (max. 1 page), a CV (including publication list), relevant diplomas, and contact details of at least 2 references.

Postdoctoral position in T cell tumor immunology in the Lillemeier laboratory at the University of Freiburg

The Lillemeier Lab is seeking a highly motivated and innovative postdoctoral fellow to investigate novel strategies for cancer immunotherapies. Our group is interested in how T cell activity and function is regulated during immune responses. Ongoing research focuses on the activating T cell receptor (TCR) and inhibitory PD-1 receptor pathways. We are identifying novel molecular mechanisms through multidisciplinary approaches that combine cellular immunology with microscopy, biophysics and biochemistry. Specifically, we have discovered that post-translational modifications control the activities of ZAP70 kinase and SHP2 phosphatase through conformational changes and receptor interaction dynamics.

The goal of this postdoctoral position is to characterize these novel mechanisms in mouse tumor models in vivo to determine if they are suitable targets for future immunotherapies. The successful candidate will investigate the effects of mutant T cell signaling molecules on anti-tumor responses using novel mouse tumor models in vivo. These tumor models have been previously established by the Lillemeier group specifically for these projects. In addition, the T cell signaling mutants will be characterized through ex vivo immunological assays and multidimensional profiling of the tumor infiltrating immune cells.

The Lillemeier lab is highly collaborative with strong ties to the immunology community in Freiburg and beyond. Thus, this postdoctoral position will be an excellent fit for a team player who is interested in collaborative research. The successful candidate will use a state-of-the-art spectral flow cytometer in the Lillemeier lab, and also have access to multiple well-equipped core facilities.

Requirements: • PhD in immunology, virology, cell biology or related field. • Publication(s) in peer-reviewed journals. • Expertise in mouse work, ideally with tumor models. A license to work with animals in the EU (e.g. FELASA certificate) is preferred. • Expertise in multi-parameter flow cytometry. If required, additional training for spectral flow cytometry will be provided. • Experience with in vitro immunological assays and tissue culture. • Familiarity with RNA sequencing and/or TCR repertoire analysis will be useful, but is not required.

This full-time postdoctoral position is available immediately (or as soon as possible) for an initial term until 31st of December 2025 with possibility of extension.

To Apply: Please include a brief letter of motivation, a description of current research accomplishments and future research interests/goals (not more than one or two pages), CV with publication list and 3+ contact details for referees in a single pdf file. Send your application in electronic form to: email hidden; JavaScript is required-freiburg.d
